2. Product Description

Alecensa (Alectinib) is a highly selective ALK (Anaplastic Lymphoma Kinase) inhibitor.
It works by blocking the activity of ALK fusion proteins, thereby preventing the growth and spread of ALK-positive cancer cells.

It is primarily indicated for ALK-positive Non-Small Cell Lung Cancer (NSCLC), including patients whose disease has progressed on or are intolerant to crizotinib.

  • First-line therapy in advanced or metastatic disease

  • For patients intolerant to or progressed on crizotinib or other ALK inhibitors

5. Side Effects

Common:

• Fatigue

• Constipation

• Muscle pain, joint pain

• Peripheral edema

• Rash, photosensitivity

• Anemia

Serious / Severe:

• Hepatotoxicity: elevated ALT/AST, bilirubin

• Interstitial lung disease (ILD)/pneumonitis: cough, dyspnea, fever

• Bradycardia: slow heart rate, dizziness, fainting

• Severe myalgia & CPK elevation

• Renal impairment (rare)

6. Precautions

• Liver Function Tests: Monitor before and during treatment.

• Creatine Phosphokinase (CPK): Check regularly; dose modifications if muscle toxicity occurs.

• Cardiac Monitoring: Monitor heart rate for bradycardia.

• Pulmonary Monitoring: Watch for ILD/pneumonitis symptoms.

• Pregnancy & Breastfeeding: Contraindicated; use effective contraception.

• Drug Interactions: Avoid strong CYP3A4 inducers/inhibitors (rifampin, carbamazepine, ketoconazole, grapefruit juice).

• Driving / Machinery: May cause dizziness or fatigue.
